Special Hemoglobin Tests

Special tests can reveal alterations in hemopglobin. The Heat Stability Test is performed for detection of unstable hemoglobins or hemoglobins with altered oxygen affinity. Some hemoglobin mutations result in an unstable hemoglobin that has decreased solubility and precipitates to form erythrocyte inclusions (Heinz bodies).  Similarly, the Isopropanol Test is performed for detection of unstable hemoglobins or hemoglobins with altered oxygen affinity. The Heinz body prep is performed in the work up for unstable hemoglobins. The Brilliant Cresyl Blue Stain tests for HbH inclusions.
